Structure
One tablet contains
active agent – pentoksifillin 600 mg,
excipients: gipromelloza, krospovidon,
microcrystalline cellulose, colloidal silicon dioxide anhydrous, magnesium stearate, macrogoal 6000, talc, titanium dioxide, polyacrylate dispersion.

Pharmacokinetics Reception properties of the tablets Vazonita retard inside provides continuous release of active agent and its uniform absorption from digestive tract.
Drug is exposed to metabolism in a liver at the first passing therefore a number pharmacological of active metabolites is formed. The maximum concentration of a pentoksifillin and its active metabolites in blood plasma is reached in 3-4 h and remains at the therapeutic level about 12 h. Drug is removed mainly with urine in the form of metabolites.
The pharmacodynamics
of Vazonit improves microcirculation and rheological properties
of blood, has vasodilating effect. As active agent contains pentoksifillin – derivative xanthine. The mechanism of action is connected with inhibition of phosphodiesterase and accumulation of tsAMF in cells of smooth muscles of vessels, in uniform elements of blood, in other fabrics and bodies. Pentoksifillin slows down aggregation of thrombocytes and erythrocytes, increases their elasticity, reduces fibrinogen level in blood plasma and strengthens a fibrinolysis that reduces viscosity of blood and improves its rheological properties. Improves supply of fabrics with oxygen in zones of the broken blood circulation, in particular, in extremities, and, to a lesser extent, in kidneys, also reduces the phenomena of a cerebral hypoxia. Slightly expands coronary vessels.

Indications
– disturbances of peripheric circulation (obliterating endarteritis, a diabetic angiopatiya, a Raynaud’s disease)
– disturbances of cerebral circulation of ischemic type (ischemic cerebral stroke)
– atherosclerotic and distsirkulyatorny encephalopathy, an angioneyropatiya
– trophic changes of fabrics owing to disturbance of arterial or venous microcirculation (posttromboflebitichesky syndrome, a varicosity, trophic ulcers, gangrene, freezing injuries)
– disturbances of blood circulation of an eye (an acute, subacute and chronic circulatory unefficiency in a retina or in an eye choroid)
– the hearing disorder of vascular genesis which is followed by relative deafness.

The route of administration and doses
Duration of treatment and the mode of dosing is set by the doctor individually, depending on a clinical picture of a disease and the gained therapeutic effect. The average duration of treatment is 2-3 weeks.
A pill of Vazonit retard should be taken inside after a meal, without chewing and washing down with enough liquid.
Drug is usually appointed on 1 tablet by 2 times a day (in the morning and in the evening).

Side effects
– nausea, loss of appetite, vomiting, diarrhea, feeling of overflow in a stomach and pain in epigastriums
– a headache, dizziness, concern, sleep disorders
– the lowering of arterial pressure, tachycardia, stenocardia, disturbances of a warm rhythm, face reddening at use of high doses
– an itching, skin rash, a small tortoiseshell, a Quincke’s edema, is rare: acute anaphylaxis.
– in rare instances: bleedings (in a GIT, in skin, in mucous) and also thrombocytopenia, aplastic anemia
– visual disturbances, scotoma, spasms.

Contraindications
– an acute myocardial infarction
– massive bleedings
– an acute hemorrhagic stroke
– an eye retinal apoplexy
– hypersensitivity to a pentoksifillin and other derivatives of methylxanthine (caffeine, theophylline, theobromine)
– pregnancy and the period of a lactation
– children’s age up to 18 years

Medicinal interactions
Pentoksifillin strengthens effect of antihypertensive drugs, anticoagulants, insulin, oral glucose-lowering drugs.

Special instructions
patients Should be careful at Vazonit’s appointment with the profound arterial hypotension, with tendency to orthostatic hypotension, in the profound coronarosclerosis and cerebral atherosclerosis with arterial hypertension, to patients with heart failure, disturbances of a warm rhythm, in digestive tract cankers, the patient who transferred surgery (risk of developing of bleedings) in a renal failure. Regular control of level of arterial blood pressure and a picture of blood
Pregnancy is recommended and a lactation
In need of use of drug in the period of a lactation it is necessary to stop feeding by a breast.
Features of influence of medicine on ability to run the vehicle or potentially dangerous mechanisms
Considering side effects of drug it is necessary to be careful at control of vehicles.

Overdose
Symptoms: nausea, dizziness, a lowering of arterial pressure, fervescence (fever), a loss of consciousness, are possible toniko-clonic spasms, vomiting a coffee thick, arrhythmias, an areflexia.
Treatment: gastric lavage, symptomatic therapy, specific antidote is absent.
