Siofor® (Metformin) oral tablets are used to treat high blood sugar levels caused by type 2 diabetes. They’re used in combination with diet and exercise. Siofor (Metformin) may be used as part of a combination therapy. That means you may need to take it with other drugs. Indications
Non-insulin dependent diabetes mellitus (type 2 diabetes) in case of ineffective diet therapy, especially if you are overweight.
Siofor® 850 can be used both as monotherapy and in combination with other oral antidiabetic agents or insulin.
Mode of application
The dosage of Siofor® 850 and the duration of use is set by the doctor for each patient individually, depending on the level of sugar in the blood and urine.
Therapy is recommended to be carried out with a gradual increase in doses, starting from 1 / 2-1 tablets of Siofor® 850 per day (which corresponds to 0.425 g - 0.850 g of metformin hydrochloride). Then, depending on the blood glucose level, the dose of the drug is increased at intervals of one week to an average daily dose of one to three Siofor® 850 tablets per day (which corresponds to 0.850 g to a maximum of 2.550 g of metformin hydrochloride per day).
The maximum single dose is 1 tablet of Siofor® 850 (0.850 g of metformin hydrochloride).
Usually, daily doses of two Siofor® 850 tablets per day are sufficient. At doses exceeding 3 Siofor® 850 tablets per day, a further increase in effectiveness is usually not achieved.
Film-coated tablets are taken without chewing and with a liquid (for example, a glass of water) during meals. If the required daily dose is more than one tablet, then it should be divided into 2 or 3 doses with meals.
Contraindications
- hypersensitivity to metformin or other components of the drug
- diabetic ketoacidosis, diabetic precoma
- severe disorders of liver and kidney function
- myocardial infarction
- cardiovascular insufficiency
- respiratory failure and hypoxic conditions
- diabetic retinopathy

Side effects
- very often (> 10%): nausea, vomiting, diarrhea, abdominal pain, loss of appetite. They appear at the beginning of treatment and in most cases go away on their own. To avoid the onset of these symptoms, it is recommended to take metformin during or after meals in 2 or 3 doses.
Discontinuation of treatment is usually not required, since in most cases these symptoms disappear even with an unchanged dosage.

- very rarely (‹0.01%): mild erythema in patients with hypersensitivity, decreased intestinal absorption of vitamin B12, as well as with long-term treatment - a decrease in its serum concentration, lactic acidosis (0.03 cases / 1000 patients per year )
Overdose
Symptoms: lactic acidosis, symptoms of which begin with nausea, vomiting, diarrhea, abdominal pain and severe weakness. Muscle pains, rapid breathing, turbidity, and loss of consciousness may occur.
Treatment: The most effective measure to remove lactate and metformin is hemodialysis. Symptomatically, measures are taken to stabilize blood circulation, level acidosis and eliminate hypoxia.
Interaction
With the simultaneous use of Siofor® 850 with other antidiabetic agents (sulfonylurea derivatives, insulin), salicylates, MAO inhibitors, oxytetracycline, ACE inhibitors, clofibrate, cyclophosphamide, the hypoglycemic effect of metformin may increase.
With simultaneous use with glucocorticosteroids, oral hormonal contraceptives, adrenaline, glucagon, thyroid hormones, phenothiazine derivatives, thiazide diuretics, nicotinic acid derivatives, the hypoglycemic effect of metformin may decrease.
