Pharmacokinetics of 45% of Phencarolum properties is quickly soaked up from digestive tract and in 30 minutes it is found in body tissues. The maximum concentration of active ingredient in blood plasma is reached in an hour. The highest concentration of active ingredient is observed in a liver, in lungs and kidneys, the lowest – in brain tissues. Phencarolum has low lipophilicity and its content in brain fabrics low (less than 0.05%) that explains lack of the oppressing influence on the central nervous system, but at individual hypersensitivity the weak sedation is possible.
Phencarolum is metabolized in a liver. Metabolites and not changed share of drug are removed generally with urine, bile and through lungs. Through intestines not absorbed part of drug is removed. The main part of drug and its metabolites (about 44%) are removed with urine within 48 hours and another 1% within the next 48 hours.

The pharmacodynamics
Phencarolum – antihistaminic means (derivative a hinuklidilkarbinola), reduces impact of a histamine on bodies and their systems. Unlike other classical drugs of this group, Phencarolum possesses the special mechanism of action, drug blocks histamine H1 receptors in peripheral fabrics and also activates enzyme diaminoxidase (histaminase), thus, reducing the maintenance of a histamine in fabrics. The efficiency of Phencarolum for patients whose treatments by other antihistaminic means inefficiently is explained by it. Phencarolum has low lipophilicity therefore badly gets through a blood-brain barrier, poorly influences processes of deamination of serotonin in tissues of a brain and on activity of enzymes of monoamine oxidase.
Phencarolum reduces toxic action of a histamine, eliminates or weakens bronkhokonstriktorny action and spazmiruyushchy influence on smooth muscles of intestines, weakens hypotensive action of a histamine and its influence on permeability of capillaries. Phencarolum possesses moderate antiserotoninovy and weak cholinolytic action, does not possess adrenolytic action.
Drug renders expressed antipruritic, desensibilizing and also moderate diuretic action.

The route of administration and doses
accept Phencarolum inside right after food.
To children up to 3 years appoint 5 mg 2 times a day, from 3 to 7 years – on 10 mg 2 times a day, from 7 to 12 years – on 10-15 mg 2-3 times a day, 12 years are more senior – on 25 mg 2-3 times a day. Duration of a course of treatment is 10-15 days. If necessary the course is repeated.

Side effects

Contraindications

Medicinal interactions
Phencarolum does not strengthen the oppressing effect of sleeping medicines on the central nervous system. Phencarolum has weak M-holinoblokiruyushchimi properties, but at decrease in motility of digestive tract the absorption of slowly absorbed medicines can amplify (for example, anticoagulants of indirect action – coumarins).

The special

Overdose
